Ritacco et al. uncover an immune-inducible pathway that explains how lysosomal damage can trigger inflammation. Priming with the cytokine interferon-γ induces the antibacterial effector protein APOL3, which, upon sensing lysosomal injury, unleashes a membranolytic attack on the cell’s own mitochondria, releasing pro-inflammatory DNA and activating innate immunity.
